目的:
了解大鼠C6胶质瘤放疗前后18F-硝基咪唑(
18
F-FMISO)PET/CT显像的特点,探讨其评估熊果酸(UA)放射增敏疗效的价值。
方法:
建立32只荷C6胶质瘤大鼠模型,按随机数字表法分成4组:A(对照)组、B(UA)组、C(放疗)组及D(UA+放疗)组,分别于放疗前及放疗后24 h进行两次显像,采集大鼠肿瘤最大标准摄取值(SUVmax-T)、同一断层对侧脊柱旁肌肉最大标准摄取值(SUVmax-M),计算两者的比值(T/M)。第2次显像结束后,免疫组化法测定肿瘤细胞乏氧诱导因子1(HIF-1)的表达情况。
结果:
A、B两组第2次显像T/M值均较首次显像显著升高(

均0.01),C、D两组放疗后显像T/M值均较放疗前显像显著降低(

=2.84、6.05,

均0.05)。放疗后第2次显像D组T/M值较C组显著降低,差异有统计学意义(

均0.01)。T/M值与HIF-1表达呈明显正相关(

结论:
UA具有对大鼠C6胶质瘤的放射增敏作用;

F-FMISO PET/CT能反映放疗前后肿瘤内部乏氧状态的改变,并 准确评估UA对大鼠C6胶质瘤的放射增敏疗效。
Objective:
To study the characteristics of
18
F-fluoromisonidazole (
18
F-FMISO) PET/CT imaging in rat C6 glioma models before and after radiotherapy
and to explore the value of 18F-FMISO PET/CT in assessing the radiosensitivity enhancement of ursolic acid (UA).
Methods:
32 rat C6 glioma models were randomly dived into four groups (8 rats in each group): group A (control group)
group B (
UA group)
group C (radiotherapy group) and group D (UA+radiotherapy group).
18
F-FMISO PET/ CT imaging was performed before radiotherapy and 24 h after radiotherapy. Tumor maximal standardized uptake value (SUVmax-T)
the lateral spinal muscle SUVmax (SUVmax-M)
the ratio of SUVmax of tumor to muscle (T/M) were calculated. All of the rats were sacrificed after radiotherapy
and pathological changes were observed by the expression of proliferation index hypoxia induciblefactor-1 (HIF-1).
Results:
At 24 h post-radiotherapy
the T/M values in groups A and B were significantly increased than that before radiotherapy (

while the T/M values in groups C and D were decreased than that before radiotherapy (

=2.84

0.05). At 24 h post-radiotherapy
the T/M value in group D was lower than that in group C
and the difference was statistically significant(

0.01). The expression of HIF-1 in group D was lower than that in other three groups (

0.01). T/M value was positively correlated with HIF-1 expression (

Conclusion:
UA could enhance the radiosensitivity of radiotherapy in treatment of rat C6 glioma. Hypoxic status within the tumor could be reflected by
18
F-FMISO PET/CT
and by which the radiosensitization effect of UA on C6 rat glioma could also be assessed.
