方法:
使用细胞计数试剂盒-8(cell counting kit-8,CCK-8)法评估Nb
0.12
-TiO
2
在无NIR激发下对人脑胶质瘤细胞(U87细胞)活性的影响;活死细胞染色评估Nb
0.12
-TiO
2
对U87细胞的光热治疗效果;实时荧光定量聚合酶链反应(real-time fluorescence quantitative polymerase chain reaction,RTFQ-PCR)评估U87细胞经Nb
0.12
-TiO
2
光热治疗后HIF-1的基因表达水平。
结果:
CCK-8法结果表明,Nb
0.12
-TiO
2
与U87细胞共培养24 h后,细胞活性>90%。活死细胞染色结果显示,在磷酸盐缓冲生理盐水(phosphate-buffered saline,PBS)组、Nb
0.12
-TiO
2
组和PBS+NIR组未出现明显的细胞凋亡,而Nb
0.12
-TiO
2
+NIR组出现大量细胞死亡。RTFQ-PCR实验发现,U87细胞经Nb
0.12
-TiO
2
+NIR治疗后,HIF-1的基因表达大幅下调,与PBS组、Nb

组和PBS+NIR组差异有统计学意义(

结论:

具有高度的生物相容性,在NIR激发下可显著杀伤U87细胞,且可同时下调HIF-1的表达,有望用于脑胶质瘤的光热治疗。
Objective:
To evaluate the photothermal therapy effects of a new types of photosensitive nanomaterial
niobium-doped titanium dioxide (Nb
0.12
-TiO
2
)
on gliomas under the excitation of near-infrared light (NIR) and the effects of Nb
0.12
-TiO
2
on hypoxia inducible factor-1 (HIF-1).
Methods:
Cell counting kit-8 (CCK-8) assays was used to evaluate the effects of Nb
0.12
-TiO
2
on the viability of human glioma cells (U87 cells) without NIR stimulation. The photothermal effect of Nb
0.12
-TiO
2
on U87 cells was evaluated by live and dead cell double staining. The gene level of HIF-1 in U87 cells after photothermal treatment with Nb
0.12
-TiO
2
was evaluated by real-time fluorescence quantitative polymerase chain reaction (RTFQ-PCR).
Results:
CCK-8 assay showed that after cocultured with Nb
0.12
-TiO
2
for 24 h
the viability of U87 cells was >90%. The results of live and dead cell double staining showed that a large number of cell death occurred in Nb
0.12
-TiO
2
+NIR group
and there was no obvious apoptosis in phosphate-buffered saline (PBS) group
Nb
0.12
-TiO
2
group and PBS+NIR group. RTFQ-PCR showed that the expression of HIF-1 was significantly downregulated in U87 cells after treated with Nb
0.12
-TiO
2
+NIR
which was significantly different from that in PBS group

Conclusion:
Nb
0.12
-TiO
2
with high biocompatibility
can significantly kill U87 cells and downregulate the expression of HIF-1 under the excitation of NIR
which is expected to be used in the photothermal treatment of glioma in future.
